Connect to The Bicho

First, make sure that you've Switch The Bicho to Programming-mode.
After that, connect The Bicho to an USB port of your computer.

Open the Car Backdoor Maker software and press the Connect button, or what is the same: go to Tools -> Connect.

Verify that the red light stays on, that means it's connected correctly.

Now, you can start with the Basic Setup and finally, Write The Bicho memory.
